Looking into the IL-1 of the storm: are inflammasomes the link between immunothrombosis and hyperinflammation in cytokine storm syndromes?

Summary Inflammasomes and the interleukin (IL)-1 family of cytokines are key mediators both inflammation immunothrombosis. responsible for release pro-inflammatory IL-1β IL-18, as well releasing tissue factor (TF), a pivotal initiator extrinsic coagulation cascade. Uncontrolled production inflammatory results in what is known “cytokine storm” leading to hyperinflammatory disease. Cytokine storms can complicate variety diseases hypercytokinemia, coagulopathies, damage, multiorgan failure, death. Patients presenting with cytokine storm syndromes have high mortality rate, driven part by disseminated intravascular (DIC). While our knowledge on factors propagating increasing, how influences DIC remains unknown, therefore treatments diseases, where these aspects feature limited, most targeting specific cytokines. Currently, no therapies target immunothrombosis aspect syndromes. Here we discuss inflammasome pyroptosis may be novel therapeutic strategy treatment hyperinflammation its associated pathologies.
